Tyrrell's was founded by Edward Tyrrell, who harvested his first vintage in 1864. Edward George "Dan" Tyrrell took over as winemaker in 1889, and Murray Tyrrell took over winemaking duties in 1959. Bruce Tyrrell joined the company in 1974 and became managing director in 2000, and Chris Tyrrell is a fifth-generation winemaker.

In 1961, the Private Bin system was created to mature fruit from vineyard blocks in individual oak barrels or vats. The first Vat 1 Hunter Semillon was produced in 1963 from the Short Flat Vineyard, and Chardonnay vines were planted in 1968, leading to Australia's first commercial Chardonnay, Vat 47 Chardonnay. In 2017, the Tyrrell family purchased the Old Hillside Vineyard, including the 1867 Old Patch block, and in 2023, the Hunter Valley Winery and Vineyards became certified members of Sustainable Winegrowing Australia.
